AAPS 2012: Investigating the Miscibility of an API in Soluplus Dispersions by Raman Chemical Imaging Technique

This poster session discusses the miscibility of drugs in the solid dispersions as it has been subject of continued interest in the development of oral dosage. The study was aimed at investigating the miscibility of carbamazepine in Soluplus matrix by using a Raman Chemical Imaging (RCI) technique.

Introduction:
Raman Chemical Imaging (RCI) spectroscopy was used to investigate the solubility of model drug Carbamazepine in solid dispersions in Soluplus, a graft polymer comprised of polyethylene glycol, polyvinylacetate and polyvinylcaprolactame. Soluplus has been used to increase the solubility and bioavailability of poorly soluble molecules.

RCI is considered one of the most powerful analytical tools for study of molecular composition, structure, identification of polymorphs and pseudo-polymorphs and other solid-state characteristics of solid solutions as it provides insight on the structure changes for each insoluble or precipitated components. Wide-field Raman Chemical Imaging (RCI) approaches image construction by assembling spatially-resolved frames into a Raman hypercube with spatial and wavelength information for each frame. Contrast at a desired wavelength of the image hypercube reveals the presence and absence of Raman scattering, and therefore, identifies the species of interest at each pixel.
